SMARTer Target RNA Capture for Illumina

SMARTer Target RNA Capture for Illumina is a complete system is designed to generate cDNA from enriched total RNA. Targeted RNA-seq can overcome several challenges inherent in whole transcriptome amplification (WTA) RNA-seq experiments, improving sequence coverage and sensitivity of detection of transcripts of interest that may be present in low amounts. Targeted RNA-seq approaches are also advantageous in saving costs and simplifying analysis. Enrichment of transcripts of interest prior to cDNA synthesis enables the capture of information about transcripts that could otherwise be missed or require a much greater number of sequencing reads to be detected, including chimeric gene fusions, transcript isoforms, and splice variants. By improving sensitivity, the power of analysis is also improved, as seen in a reduction in the signal-to-noise ratio.

SMARTer Target RNA Capture for Illumina is used to generate cDNA from enriched RNA transcripts. Target-specific biotinylated DNA probes designed by the user are hybridized to transcripts of interest and the resulting RNA-DNA hybrids are captured with Capture Beads. This kit takes advantage of the sensitivity of the SMART-Seq® v4 Ultra® Low Input RNA Kit for Sequencing to generate high-quality, full-length cDNA from these enriched transcripts. The kit was tested with 10 ng of total RNA, and has been shown to perform cDNA synthesis from enriched transcripts derived from as little as 100 pg purified total RNA. cDNA libraries generated by this kit have been tested for compatibility with Illumina sequencing platforms. This method does not require additional rRNA removal methods or kits.

Capture Beads are magnetic particles with streptavidin coated on the surface. They exhibit very low, non-specific adsorption of protein and nucleic acids on the particle surface, and therefore can be used for recovering high-purity, biotin-labeled molecules from samples. Additionally, the hydrophilic polymer on the Capture Beads does not inhibit enzymatic reactions such as PCR or reverse transcription. Capture Beads are used as a part of the components set for the SMARTer® Target RNA Capture for Illumina® kit.
