SMART-Seq HT Kit

The SMART-Seq HT Kit uses oligo dT priming to generate high-quality, full-length cDNA directly from 1–100 cells or 10 pg–1 ng of total RNA with a streamlined protocol that is optimized to work downstream of FACS. This kit features a simplified protocol that reduces hands-on time compared to SMART-Seq v4, including the introduction of a convenient combined reverse transcription and PCR amplification step. Despite the changes to the protocol, SMART-Seq HT delivers the same high sensitivity and reproducibility as SMART-Seq v4.

The SMART-Seq HT Kit is designed for synthesis of high-quality cDNA directly from 1–100 intact cells or ultra-low amounts of total RNA (10–1,000 pg). The simplified workflow of this kit allows users to process a large number of samples with ease. cDNA libraries generated with this kit have been tested for compatibility with Illumina sequencing platforms.

The SMART-Seq HT Lysis Components are accessories to the SMART-Seq HT Kit. These components consist of 10X Lysis Buffer as well as the 3′ SMART-Seq CDS Primer II A, and are provided in separate packaging for users who want to use FACS to sort a large number of cells or nuclei without using all of the sorted material as input for cDNA synthesis with the SMART-Seq HT Kit.
