Detailed separation procedure
CD4+ T cells are isolated by depletion of non CD4+ T cells using a cocktail of biotin-conjugated antibodies against CD8a, CD11b, CD11c, CD19, CD45R (B220), CD49b (DX5), CD105, Anti-MHC-class II, Ter-119 and TCRγ/δ as primary labeling reagent. The cells are then magnetically labeled with Anti-Biotin MicroBeads. The magnetically labeled non-target cells are depleted by retaining them on a MACS® Column in the magnetic field of a MACS Separator, while the unlabeled target cells pass through the column.
Downstream applications
The CD4+ T Cell Isolation Kit, mouse is used where untouched CD4+ T cells are desired or where further magnetic separation into T cell subsets is required.
