Whole-genome amplification

The PicoPLEX WGA family of products is comprised of three versions. All of the systems below can amplify DNA directly from cell lysates from 1–10 cells and can also be used with isolated gDNA inputs ranging from less than 6 pg up to 50 pg. They are intended for use with array CGH, qPCR, and NGS applications.

Overview

  • Reproducible results from single cells (or 6–50 pg of DNA)
  • Superior reproducibility of allele representation
  • Easy to use and automate
  • Unambiguous results at all resolutions (e.g., microarray and PCR)

Applications

  • CNV analysis using NGS, oligonucleotide aCGH, BAC aCGH, and qPCR
  • Target enrichment or amplicon enrichment for mutation detection
  • SNP genotyping

Suitable Inputs

  • Mammalian cells (1–10 cells)
  • Bacterial cells (1,000–10,000 cells)
  • Isolated DNA (6–50 pg of human DNA)
  • Sorted chromosomes
  • Intact or fragmented, single- or double-stranded DNA

The PicoPLEX Single Cell WGA Kit uses a single-tube protocol developed specifically for reproducible amplification of single-copy genomic DNA (gDNA) starting with 1–10 cells or equivalent picogram quantities of isolated gDNA. Cell lysis and whole-genome preamplification are followed by amplification, which results in libraries with very low background and typical yields of 8–12 µg of product in under 3 hours. This system is best suited for applications in which high yield is most important. This product contains reagents for 24 reactions.

The PicoPLEX WGA Kit uses a single-tube protocol developed specifically for the reproducible amplification of genomic DNA (gDNA) starting from 1–10 cells or equivalent picogram quantities of isolated gDNA. Cell lysis and whole-genome preamplification are followed by amplification with very low background to yield over 2 µg of product in under 3 hours. This is the kit of choice for assays that need the most accurate and reproducible measurement of CNV. This product contains reagents for 50 reactions.
